Description

Fluorescent in situ hybridization (FISH) data enumerated from microscopic samples on 2 legs of each of the CAR212 and CAR216 cruises.

Methods & Sampling

All samples were collected via Niskin bottles.

Seawater samples were fixed with 2% borate-buffered formaldehyde, frozen overnight, then filtered the next day onto 0.2-um polycarbonate membrane filters. Filters were refrozen at -20 degrees celsius until hybridization. All FISH hybridization procedures were performed according to Pernthaler et al. (2001) with fluorescent probes labeled with CY3 or fluorescein. Filters were counterstained with DAPI and probe-positive cells were enumerated as a percentage of the total community (DAPI-positive). All data were normalized to volume of seawater.

Generic Description:

A Niskin bottle (a next generation water sampler based on the Nansen bottle) is a cylindrical, non-metallic water collection device with stoppers at both ends. The bottles can be attached individually on a hydrowire or deployed in 12, 24, or 36 bottle Rosette systems mounted on a frame and combined with a CTD. Niskin bottles are used to collect discrete water samples for a range of measurements including pigments, nutrients, plankton, etc.
